In this scientific report, some of the cellular and molecular effects underlying the observation that the snail secretions have regenerative effects on human skin are outlined. First, the secretion has enzyme antioxidant SOD (Superoxide Dismutase) and GST (Glutathione - often called the "master antioxidant") activities, as well as multiple modes of antioxidant action, acting at the level of free radical production and also sequestering free radicals. In addition, it stimulates fibroblast proliferation and rearrangement of the actin cytoskeleton of cells.

Additional mechanisms involved in the regenerative effect of the secretion include the stimulation of extracellular matrix assembly and the action of regulatory proteins known as Tissue Inhibitors of Metalloproteinases (TIMPs) responsible for the reversible inhibition of metalloproteinase (MMP) enzymatic activities, responsible for maintaining a balance between the degrading of damaged tissues and their new synthesis. This also limits the extent of the damage during wounding and scar formation and attenuates the signs of aging by reducing the degenerative effects of what usually becomes excessive MMP enzymatic activity.

Ultraviolet light as well as frequent contact with harmful environmental chemicals such as ozone leads to the formation of excessive quantities of reactive oxygen species on the skin. Free reactive oxygen radicals cause destruction of biomembranes by oxidizing unsaturated fatty acids and protein oxidation in human as well as depolarization and crosslinking of biopolymers. This induces a spectrum of well documented acute responses such as erythema, hyperproliferation, desquamation and with chronic irritation a general degradation of collagen together with an accumulation of abnormal elastin deposits in the dermis resulting in premature skin aging.

The skin's own defense system is mainly based on the functionality of superoxide dismutase (SOD). Endogenic SOD is found at high levels in the stratum corneum but only low baseline levels are found in the dermis of people with photoaged skin. Also a significant depletion of antioxidants enzyme expression is observed within the strateum corneum and in their epidermis. The endogenous level of SOD decreases significantly. A replacement by using cosmetic formulations is therefore necessary. Externally applied SOD is able to penetrate into the intact stratum corneum and reach the upper layers of the epidermis, as has been shown using a fluorescent dye (FITC) labeled enzyme.

The report confirms a protective effect of the snail secretions in topical application against UV induced cutaneous damage and to overcome a lack of endogenic SOD necessary to restore the antioxidative homeostasis of the skin.

A cream formulated with the snail serum was applied to the face of 32 individuals who showed signs of facial photoaging for an eight-week period on a continuous basis. The subjects were evaluated at 2, 4 and 8 weeks of treatment using a clinical evaluation method and the results were correlated with those seen with the use of instrumental techniques that measured wrinkle depth, smoothness, hydration, firmness and elasticity.

Subjects also kept a subjective journal of their own observations and feelings towards the product which were evaluated at the end of the study.
